The Janssen R&D Discovery, Product Development, & Supply (DPDS) Postdoctoral Fellowship Program helps early career scientists gain the training and experience needed to further their careers, while addressing the needs of global healthcare. BDDS (Bioanalytical Discovery and Development Sciences) is a critical part of Janssen R&D and is committed to the delivery of key data for understanding of pharmacokinetics, toxicokinetics, pharmacodynamics, and immunogenicity across Janssen R&D’s entire portfolio and at all stages of drug discovery and development. Lipids are far from inert molecules and exert critical signaling and cellular functions that are at the center of many metabolic diseases such as NASH or neurological disorders such as Parkinson’s disease. Lipid remodeling can occur as a consequence of disease progression or resolution, and lipid signatures are examined as biomarkers for drug intervention. It is critical for BDDS to develop advanced lipidomics analytical tools to enhance our capability to assess complex lipid metabolism and remodeling for identification of novel drug targets or biomarkers. Development of such lipidomics analysis platform is critical for the success for several Janssen therapeutic areas such as cardiovascular and metabolic disease (CVM) and neuroscience (NS).
The aim of the project is to develop cutting-edge analytical tools and answer challenging biological questions related to lipid remodeling and fatty acid trafficking in pre-clinical disease models in metabolism and neuroscience. The candidate will utilize mass spectrometry-based techniques to study inter- and intra- lipid fatty acid trafficking to understand the mechanisms underlying various disease states, and also identify novel biomarkers of disease progression or therapeutic efficacy for novel drug candidates. The candidate will also develop a comprehensive quantitative targeted lipidomics platform to evaluate global lipid changes, and novel methodologies such as lipid class separation by solid phase extraction prior to LC-MS/MS or LC-HRMS analysis to study lipid remodeling in response to disease and/or therapeutic intervention.
The Postdoctoral Fellow will work closely with our therapeutic areas to apply those methodologies to preclinical disease models and compare to human disease signatures to help uncover novel biology and biomarkers for ongoing programs. Furthermore, the candidate will have the opportunity to evaluate novel commercially available methodologies such as electron activated dissociation fragmentation for their effectiveness in supporting the above objectives. The candidate will be part of a dynamic team and work closely with colleagues in Janssen’s global bioanalytical teams, as well as colleagues associated with different therapeutic areas.
The candidate’s responsibilities will include:
- Development and validation of lipid remodeling profiling method
- Application of lipid remodeling profiling method to cardiovascular metabolism therapeutic area
- Application of lipid remodeling profiling method to neuroscience therapeutic area
- Publication writing
- Development, training, conferences, internal meetings & symposia
Qualifications
Required Minimum Education
- PhD Required Years of Related Experience: 0-1
Required Knowledge, Skills, and Abilities:
- Hands-on experience with metabolomics or lipidomics analysis by LC-MS
- Hands-on experience with quantitative bioanalysis in complex biological matrices such as plasma, serum, and tissue
- Hands-on experience with bioanalytical sample preparation skills, such as solid phase extraction, liquid-liquid extraction, and protein precipitation
- Knowledge of lipid biochemistry, particularly related to cardiovascular and metabolic diseases
Preferred Area of Study:
Biological, biochemistry, pharmacology, analytical chemistry, or related field
Preferred Knowledge, Skills and Abilities:
- Experience with software and tools for multivariate statistical analysis
- Hands-on experience with advanced chromatography techniques such as capillary-LC or nano-LC
- Knowledge of Absorption, Distribution, Metabolism, and Excretion (ADME) and pharmacokinetics
